The young Magpies, who finished in 21st place of the Premier League 2, make the long trip to South Wales as they take on their Swans counterparts at the Swansea.com Stadium [kick-off 7pm BST], targeting a place in the quarter-final stages.
Charlton, who signed his first professional contract with United in the summer of 2024, is determined for Newcastle's second-string to complete a long passage in the newly-introduced play-offs, which involves the sides finishing between 17th and 24th place as well as the top eight PDL sides.
The teenage left-back told newcastleunited.com: "It's another big game for us, a knockout game, and we want to go as far as possible before the end of the season.
"The further you go into the play-offs, the harder the games get. We always want to play against the best teams. It's going to be a different challenge playing against a Category 2 side because they're not used to playing against those teams.
"We'll expect them to be very physical and technical. It's a huge club and a really good stadium. Playing at a first-team ground brings around a good feeling for this game.
"We're on the right track at this moment in time. Robbie [Stockdale] has came in and the form has been better as of late, getting strong results but we know we will need to deliver a strong game to go through.
"We want to go as far as possible. We've beaten the likes of Man United, Aston Villa and went toe-to-toe with Arsenal so we're more than capable of winning the competition."
The Gateshead-born youngster recently marked his return from a hamstring injury sustained in February, playing the opening hour of Newcastle's 3-1 away win against Manchester United.
Charlton, who joined Newcastle's Academy as a 13-year-old, reflected on a difficult spell on the sidelines and is aiming to gain more minutes before the U21s' conclusion of the 2025/26 season.
"I had a tear on my hamstring which was tough to take," he explained. "I didn't know if I was injured or not before getting a scan and being told I would be out for two months.
"I completed the rehab and was able to play against Man United, getting some important minutes, and we won the game which was great. "Everyone wants to make an impact for the team whether you're playing or not. As a group, we want to finish the season as strongly as possible and be able to play as many matches as we can."
